If all of my opponents (or perhaps just one) are unable to be targetted, does that mean Descendant of Masumaro never loses counters?

only if it's all of them. you have to choose a valid target if one exists.

Important note: The entire ability will do nothing if none of your opponents can be targeted. It just won't go on the stack.
